چکیده
The front cover picture shows a prefluorescent phosphine-based probe reacting with nitroxyl (HNO) and S-nitrosothiol (RSNO), nitrogen oxide-derived biological signals. Both species react with the prefluorescent probe, but only the product from the HNO reaction can complete a further chemical ligation pathway that results in fluorescence, indicating the presence of HNO. The product of the probe with RSNO does not complete this ligation and does not generate a fluorescent species. These phosphine-based probes thus demonstrate a selectivity for HNO over RSNO based on their chemical reactivity and can be used in biological systems to differentiate these species. More information can be found in the Communication by S. Bruce King and Z. Miao on page 110 in Issue 2, 2016 (DOI: 10.1002/open.201500200).

Comparison of Reductive Ligation‐Based Detection Strategies for Nitroxyl (HNO) and S‐Nitrosothiols
Phosphine-based detection strategies for both nitroxyl (HNO) and S-nitrosothiols (RSNO) were investigated and compared. Phosphorus NMR studies show that azaylides derived from HNO or organic RSNO efficiently participate in subsequent reductive ligation required for fluorescence generation in properly substituted substrates. S-Azaylides derived from biological RSNO containing free amine and carb...

A reductive ligation based fluorescent probe () for the detection of S-nitrosothiols (SNO) was developed. The probe showed good selectivity and sensitivity for SNO.
متن کاملNitroxyl (HNO): A Reduced Form of Nitric Oxide with Distinct Chemical, Pharmacological, and Therapeutic Properties
Nitroxyl (HNO), the one-electron reduced form of nitric oxide (NO), shows a distinct chemical and biological profile from that of NO. HNO is currently being viewed as a vasodilator and positive inotropic agent that can be used as a potential treatment for heart failure. The ability of HNO to react with thiols and thiol containing proteins is largely used to explain the possible biological actio...
متن کاملA highly sensitive and reductant-resistant fluorescent probe for nitroxyl in aqueous solution and serum.
A novel coumarin-based fluorescent probe, P-CM, for quantitative detection of nitroxyl (HNO) was developed. P-CM exhibits a selective response to HNO over other biological reductants and was also applied for quantitative detection of HNO in bovine serum with satisfactory results.
متن کاملVisualization of nitroxyl (HNO) in vivo via a lysosome-targetable near-infrared fluorescent probe.
We have presented a near-infrared fluorescent probe Lyso-JN for the detection of nitroxyl (HNO) in cells and in vivo. Lyso-JN is comprised of three moieties: an Aza-BODIPY fluorophore, a HNO-response modulator, diphenylphosphino-benzoyl, and a lysosomal locator, alkylmorpholine. The detection mechanism is based on aza-ylide intramolecular ester aminolysis reaction with HNO. The probe holds the ...
